MCF10A cells transfected with empty vector were grown on cover slips, washed with PBS, and fixed with methanol/acetone (1:1, v/v) before incubation with mouse anti-β-catenin antibody. The β-catenin protein was detected by incubation with Texas Red-conjugated anti-mouse IgG antibody. Coverslips were covered with antifade solution containing DAPI and viewed by confocal microscopy. Immunofluorescence staining demonstrated that majority of the β-catenin protein is localized on the membranes. For details, see article by Shekhar et al. on page 729.
