HaCaT keratinocyte cells were transiently transfected with a plasmid encoding ZACD2, a variant of the candidate tumor suppressor protein ZAC. ZACD2 protein was detected using a rabbit anti-ZAC antibody followed by anti-rabbit Cy3 (red-orange). Involucrin was detected using a mouse anti-involucrin monoclonal antibody followed by anti-mouse FITC (green). Cells were counterstained with DAPI (blue). A ZACD2-negative cell on the left (blue nucleus) displays only basal involucrin staining. In contrast, the ZACD2-positive cell on the right (pink nucleus) shows abundant involucrin expression, indicating that ZACD2 expression leads to keratinocyte differentiation.
