Rat-1 fibroblasts were cultured on glass coverslips, incubated for 6 hours with 10 mM sodium butyrate, fixed in cold methanol, and immunofluorescently labeled with rabbit antibodies against cingulin and mouse antibodies against ZO-1, followed by Cy5-labeled (red) anti-rabbit and FITClabeled (green) anti-mouse secondary antibodies. The mounting medium contained DAPI, which stains cell nuclei (blue). Sodium butyrate induces up-regulation of the expression of cingulin, which co-localizes with ZO-1 at cell-cell junctions (yellow).
