Hypoxic cell populations (yellow/orange) in frozen sections from a human breast cancer xenograft overexpressing HER-2/neu (MDA-435/LCC6HER-2) visualized using immunofluorescence. Mice were exposed to 2-(2-nitro-1H-imidazol-1-yl)-N-(2,2,3,3,3- pentafluoropropyl)-acetamide (EF5) before tumor harvest. EF5 forms adducts to cellular proteins in viable hypoxic cells that are subsequently detected with anti-EF5 antibody conjugated to CY3. Sections were then counterstained with DAPl (blue). Images were acquired separately for each fluorochrome and merged using Adobe Photoshop. For details, see the article by Dragowska et al. on page 606.
